Uniwersalny take a look at immunologiczny ze znacznikiem SERS do wykrywania chorób chorobotwórczych w obniżeniu ceny o separację Fe   O   @ Au-aptamer i rozpoznawanie orientacji przeciwciało-białko A

 

Pożądane są szybkie, niezawodne i czułe metody wykrywania bakterii chorobotwórczych. W niniejszym dokumencie zaproponowaliśmy take a look at immunologiczny ze wspomaganym magnetycznie wspomaganym powierzchniowo rozpraszaniem Ramana (SERS) do czułego wykrywania bakterii przy użyciu uniwersalnego podejścia opartego na  znakowaniu wolnych  przeciwciał i rozpoznawaniu orientacji znaczników białek gronkowca A (PA) -SERS. Biosensor SERS składa się z dwóch funkcjonalnych nanomateriałów: sprzężonych z aptamerem nanocząstek magnetycznych  Fe 3 O 4 @Au (MNP) jako platformy magnetycznej SERS do wzbogacania patogenów oraz znaczników SERS zmodyfikowanych PA (Au @ DTNB @ PA) jako uniwersalnej sondy dla bakterii docelowych wykrywanie ilościowe. Po wzbogaceniu bakterii docelowych, wolne  przeciwciało został użyty do specyficznego znakowania bakterii docelowych i dostarczył licznych fragmentów Fc, które mogą kierować wiązaniem zależnym od orientacji znaczników PA-SERS.

Dzięki tej strategii, znaczniki Fe 3 O 4 @ Au / bakterie / SERS oznaczały wielowarstwowe kompleksy immunologiczne dla większości bakterii (spodziewając się kilku gatunków Staphylococcus). Stwierdzono, że granice wykrywalności (LOD) proponowanego testu wyniosły 10, 10 i 25 komórek / ml dla trzech powszechnych patogenów Escherichia coli (E. coli), Listeria monocytogenes (L. mono) i Salmonella typhimurium (S. typhi) w prawdziwych próbkach żywności. Uniwersalna metoda wykazuje również zalety szybkiej, solidnej i łatwej w obsłudze, co sugeruje jej duży potencjał w zakresie monitorowania bezpieczeństwa żywności i diagnostyki chorób zakaźnych.

Wzmocnienie wydajności poprzez tworzenie wzoru produkcji gradientu, zakładanie immunosensora elektrochemiluminescencyjnego, tworzenie za pomocą magnetolitografii w oznaczaniu albuminy surowicy ludzkiej

 

Właściwości gradientu ułatwiają korelację właściwości chemicznych i fizycznych cząstek na strukturę i przylegający los. W tym przypadku poprawę wydajności bada się za pomocą wzorcowania gradientu cząstek (PGAP) utworzonego z gradientem pola magnetycznego (MFG) metodą magnetolitografii (ML) w pomiarze elektrochemiluminescencji (ECL). Magnetyczne  nanocząsteczki Fe 3 O 4 zostały wybrane jako nanonośniki i pokryte warstwą SiO 2 w  celu unieruchomienia przeciwciał pierwotnych  . Kropki kwantowe CdTe z powłokami z białka G wybrano jako znaczniki sygnałowe i  skoniugowano  z przeciwciałami drugorzędowymi . Namagnesowany słupek 500 nm, matryce niklu z linią 1 μm i Three μm umieszczono za elektrodą roboczą modyfikując immunosensor ECL o strukturze warstwowej w celu utworzenia różnych PGAP.

Zwiększenie wydajności o okay. 2,Four raza zaobserwowano porównując immunosensor wolny od PGAP z badanym immunosensorem gradientowym , utworzonym z namagnesowanej linii niklu o grubości Three μm. Wynika z tego, że czułość immunosensora ECL została zwiększona dzięki właściwościom PGAP. Gdy immunosensor o właściwościach PGAP został użyty do ilościowego oznaczenia albuminy surowicy ludzkiej, wykazał szeroki zakres liniowy (10-480 ng / ml) i granicę wykrywalności 10 ng / ml. Właściwości PGAP, utworzone z MFG przez ML, zapewniają prostą metodę poprawy wydajności ECL.

Sprzęganie przeciwciała z polimerem wrażliwym na temperaturę  poprzez reakcję typu click on in situ  w celu umożliwienia wzbogacenia biomarkerem w celu testu czułości diagnostycznej

 

Wczesna diagnostyka chorób zakaźnych jest jednym z obecnie powszechnych wyzwań, zwłaszcza w środowiskach o niskich i ograniczonych zasobach, gdzie potrzebne są proste, szybkie, przenośne, tanie i czułe metody diagnostyczne. Check immunologiczny przepływu bocznego (LFIA) jest powszechnym, szybkim testem przesiewowym. Jednak niska czułość testu ogranicza użyteczność LFIA w przypadku próbek o małej liczbie patogenów (wczesne stadia infekcji). Przeciwciała  sprzężone  z polimerami reagującymi na bodziec były wcześniej wykorzystywane do poprawy czułości testów do wykrywania biomarkerów w niskich stężeniach. Jednak utrata  przeciwciał powinowactwo po koniugacji polimeru pozostaje znaczącym wyzwaniem. W tym badaniu opracowaliśmy poli (N-izopropyloakrylamid-ko-N- (2-hydroksyizopropylo) akryloamid-szczepiony wspólnie alkinem izopropyloakryloamidem), nowy polimer do wzbogacania biomarkerów, poprzez koniugację polimeru po  rozpoznaniu przeciwciała i antygenu.

Zastosowaliśmy i promowaliśmy chemię kliknięć in situ, aby ułatwić wysoce specyficzną koniugację między nowymi polimerami wrażliwymi na temperaturę i kompleksami przeciwciało –  antygen . Ta metoda może tłumić spadek stałej wiązania związany z koniugacją polimeru (> 20-krotny). Koniugację z powodzeniem wykazano w płynach ustrojowych, takich jak mocz i ślina. Osiągnęliśmy> 5-krotne wzbogacenie antygenu poprzez wytrącanie termiczne poprzez sprzęganie polimerów z  przeciwciałami  po rozpoznaniu antygenu. Skoncentrowane biomarkery zaowocowały ulepszonym wykrywaniem LFIA. Podejście to można potencjalnie wykorzystać do ulepszenia testów diagnostycznych w zakresie chorób zakaźnych w warunkach niskich i ograniczonych zasobów.

Błąd połączenia.

Ocena Cys (ATTO 488) Eight Dermorphin-NH2 jako nowe narzędzia do badania receptorów peptydowych opioidowych μ

 

Receptor peptydu opioidowego μ (MOP) należy do rodziny receptorów opioidowych i jest ważnym celem klinicznym analgezji. Do pomiaru lokalizacji receptora MOP i śledzenia jego obrotu tradycyjnie używano radioiznaków lub  przeciwciał  z towarzyszącymi problemami użyteczności radioizotopów w całych komórkach i słabą   selektywnością przeciwciał . Aby rozwiązać te problemy, zsyntetyzowaliśmy i scharakteryzowaliśmy nowy fluorescencyjny analog dermorfiny oparty na ATTO488; [Cys (ATTO 488) 8] Dermorfina-NH2 (DermATTO488). Początkowo oceniliśmy profil wiązania DermATTO488 w komórkach HEK eksprymujących ludzkie komórki MOP i CHO wyrażające ludzki MOP, peptyd opioidowy δ (DOP), peptyd opioidowy κ (KOP) i peptyd Nocyceptyna / Orphanina FQ (NOP) przy użyciu wiązania radioligandu. Funkcjonalna aktywność  sprzężonego peptyd oceniano przez pomiar (i) zdolności liganda do angażowania białka G przez pomiar zdolności do stymulowania wiązania GTPγ [35S] i (ii) zdolności do stymulowania fosforylacji ERK1 / 2. Umiejscowienie receptora uwidoczniono za pomocą konfokalnej skaningowej mikroskopii laserowej.

Dermorfina i DermATTO488 wiązały się z HEKMOP (pKi: 8,29 i 7,00; p <0,05), CHOMOP (pKi: 9,26 i 8,12; p <0,05) i CHODOP (pKi: 7,03 i 7,16; p> 0,05). Oba ligandy były nieaktywne w KOP i NOP. Dermorfina i DermATTO488 stymulowały wiązanie GTPγ [35S] z podobnymi wartościami pEC50 (7,84 i 7,62; p> 0,05) i Emax (1,52 i 1,34-krotność p> 0,05). Ponadto Dermorphin i DermATTO488 wytworzyły jednofazową stymulację fosforylacji ERK1 / 2, osiągając szczyt po 5 minutach (6,98 i 7,64 razy; p> 0,05). Wreszcie, w mikroskopii konfokalnej DermATTO488 wiązał się z rekombinowanymi receptorami MOP na komórkach CHO i HEK w sposób zależny od stężenia, który można było zablokować przez wstępną inkubację z nieznakowaną dermorfiną lub naloksonem. Łącznie oprócz ATTO488Dermorphin wytworzył ligand podobny do Dermorphin; z ~ 10-krotną selektywnością w stosunku do DOP. Ten nowy ligand DermATTO488 zachował aktywność funkcjonalną i może być użyty do wizualizacji lokalizacji receptora MOP.

Metoda selektywnej koniugacji N-końca koryerza okno terapeutyczne koniugatów przeciwciało-lek poprzez poprawę tolerancji i sprzedaży

 

Koniugaty przeciwciało- lek (ADC) to ukierunkowane środki terapeutyczne, które leczą nowotwory poprzez selektywne dostarczanie wysoce silnych leków cytotoksycznych do komórek nowotworowych za pośrednictwem przeciwciał specyficznych dla raka . Jednak ich korzyści kliniczne są ograniczone przez toksyczność poza celem i wąskie okno terapeutyczne. Aby przezwyciężyć te ograniczenia, zastosowaliśmy redukcyjne alkilowanie, aby opracować nowy typ ADC, który ma leki cytotoksyczne  sprzężone  z N-końcem  przeciwciała  poprzez wiązania aminowe wprowadzone poprzez reakcje redukcyjnego alkilowania (NTERM). Aby sprawdzić, czy sprzężony NTERM ADC mogą poszerzyć zakres terapeutyczny, zsyntetyzowaliśmy trzy różne ADC przez sprzęganie trastuzumabu i monometyloaurystatyny-F trzema różnymi metodami i porównaliśmy ich stabilność, skuteczność i toksyczność.

NTERM- sprzężone  ADC bardziej stabilny  in vitro  i  in vivo  niż tiolowych sprzężone  i z cZ sprzężonych  przetworników analogowo-cyfrowych.  ADC skoniugowany z NTERM wykazał mniejszą toksyczność w porównaniu z innymi ADC, podczas gdy jego skuteczność była porównywalna z  ADC skoniugowanego z tiolem i lepsza niż  ADC skoniugowanego z lizyną. Wyniki te sugerują, że metoda koniugacji NTERM może poszerzyć okno terapeutyczne ADC poprzez zwiększenie ich stabilności i zmniejszenie toksyczności.
