ICELISA na przeciwciało monoklonalne do badania przesiewowego

Strukturalny wpływ glikodendrymerów rekrutujących przeciwciała (ARG) na cytotoksyczność przeciwnowotworową

 

Rekrutacja endogennych  przeciwciał  przeciwko komórkom rakowym stała się niezawodną przeciwnowotworową alternatywą immunoterapeutyczną w ciągu ostatniej dekady.  Rzeczywiście wykazano, że kowalencyjne przyłączenie  przeciwciał i modułów wiążących nowotwór (ABM i TBM) w jednej, dobrze zdefiniowanej syntetycznej cząsteczce sprzyja tworzeniu się oddziałującego trójskładnikowego kompleksu między  przeciwciałami  a komórką docelową, co zwykle skutkuje jednoczesnym zniszczenie komórek za pośrednictwem układu odpornościowego. We wstępnym badaniu opisaliśmy pierwsze  przeciwciało Rekrutacja glikodendrymerów (ARG), łączenie cRGD jako ligandów dla linii komórek czerniaka M21 z ekspresją αVβ3 i Rha jako ligandu dla naturalnej IgM i wykazała, że ​​wielowartościowość jest niezbędnym wymogiem do utworzenia tego kompleksu. W niniejszym badaniu zsyntetyzowaliśmy nową serię ARG składających się z ABM, tj. Samoskondensowanych ramnozylowanych cyklopeptydów i dendrymerów polilizynowych, które zostały  sprzężone  z TBM z lub bez odstępnika.

Doświadczenia z cytometrią przepływową i mikroskopią konfokalną z ludzką surowicą i różnymi liniami komórkowymi ujawniły, że geometria ABM znacząco wpływa na tworzenie trójskładnikowego kompleksu w M21, podczas gdy w BT 549 o niskiej ekspresji integryny nie występuje żadne znaczące wiązanie . Ponadto wykazujemy za pomocą testu żywotności komórek, że ARG indukują wysoki poziom cytotoksyczności wobec M21, co jest również w ścisłej korelacji ze strukturą ABM. W szczególności wykazaliśmy, że ARG łączący rdzeń cyklopeptydowy i rozgałęzienia, z lub bez przerywnika, indukuje 40-57% selektywnej cytotoksyczności wobec komórek M21 w obecności ludzkiej surowicy jako wyjątkowego źródła efektorów odporności. Na koniec podkreślamy również, że odstęp między ABM i TBM umożliwia zwiększenie cytotoksyczności pośredniczonej przez układ odpornościowy nawet przy ABM o niższej wartościowości.

Description: CD40 (48 to 50 kDa) is a transmembrane glycoprotein mainly expressed on the surface of B cells and also expressed on monocytes, dendritic cells, and thymic epithelium. CD40 is a member of the tumor necrosis factor (TNF) receptor superfamily, which includes the low affinity nerve growth factor (NGF) receptor and CD95/Fas. CD40 is the receptor for CD40 ligand. ICELISA na przeciwciało monoklonalne do badania przesiewowego diazynonu w próbkach warzyw

 

Diazynon (DAZ) to pestycyd fosforoorganiczny (OP), który jest powszechnie stosowany do zapobiegania i zwalczania szkodników zagrażających produktom rolnym. W tym badaniu opracowaliśmy nowatorską strategię powlekania heterologicznego do testu immunologicznego DAZ.

Hapten powlekający DAZ może być bezpośrednio  sprzężony  z białkiem nośnikowym bez konieczności stosowania ramienia dystansującego. Ta proponowana strategia powlekania haptenem oszczędza czas i znacznie poprawia czułość testu immunologicznego z powodu braku ramienia rozdzielającego. Zsyntetyzowany antygen powlekający w postaci zsyntetyzowanej zastosowano do skriningu przeciwciała monoklonalnego   (mAb).

Wreszcie rozwinięty pośredni konkurencyjny check immunoenzymatyczny (icELISA) wykazała układu scalonego 50  , a granica wykrywalności (LOD) wartości 0,58 ng ml -1  i Eight str ml -1 , odpowiednio. Metoda ta wykazywała pomijalną reaktywność krzyżową z innymi analogami, a odzysk próbek (ogórek, kapusta i sałata) wahał się od 92,6% do 125,4%, ze współczynnikami wariancji (CV) poniżej 12%.

Uzyskano dobrą korelację między icELISA a wysokosprawną chromatografią cieczową ze spektrometrią mas (HPLC-MS / MS). Proponowany icELISA był idealnym narzędziem do monitorowania pozostałości DAZ w próbkach żywności.

Odpowiedni środek utrwalający do barwienia MEM-G / 9 hodowanych komórek komórek nowotworowych trofoblastów JEG-Three HLA-G-dodatnich

 

Ludzki antygen leukocytów (HLA-G) uczestniczy w immunosupresji i jest przydatny w diagnostyce prenatalnej.  Podstawą nieinwazyjnych badań prenatalnych jest izolacja komórek płodowych dodatnich pod kątem HLA-G przez skoniugowane z przeciwciałem  HLA-G  nanocząsteczki z szyjki macicy kobiet w ciąży. Próbki szyjki macicy są utrwalane w pożywce transportowej przed izolacją przy użyciu   nanocząstek sprzężonych z przeciwciałami .

Barwienie HLA-G przy użyciu przeciwciała MEM-G / 9 jest  jednak ograniczone do nieutrwalonych komórek. Zbadaliśmy wpływ kilku środków utrwalających na interakcję HLA-G z MEM-G / 9 w linii komórkowej HLA-G-dodatniej JEG-3. Zbadaliśmy absolutny metanol, bufor octanowy 1: 1: metanol, roztwór Pap i paraformaldehyd.

Efekty tych utrwalaczy oceniano za pomocą immunofluorescencji. Nie stwierdzono barwienia powierzchni MEM-G / 9 komórek utrwalonych metanolem. Około 40% komórek JEG-Three utrwalonych paraformaldehydem nie wybarwiło się.

Prawie wszystkie komórki barwiono MEM-G / 9 po utrwaleniu buforem octanowym: metanolem lub roztworem Pap. Nasze odkrycia wskazują na znaczenie stosowania odpowiedniego utrwalacza do konserwacji antygenu powierzchniowego komórek HLA-G do badań z użyciem przeciwciała MEM-G / 9  .

Produkcja w Escherichia coli rekombinowanych fragmentów białek wypustek COVID-19 połączonych z CRM197

W 2020 roku pandemia COVID-19 dotknęła prawie 10 8  osób. W związku z tym opracowano całkiem sporo szczepionek przeciwko COVID-19, a kilka ostatnio otrzymało zezwolenie na użycie w nagłych wypadkach. Ogólnie rzecz biorąc, szczepionki te są ukierunkowane na określone białka wirusowe przez  przeciwciała,  których synteza jest bezpośrednio lub pośrednio wyzwalana przez kwasy nukleinowe kodujące pożądane cele.

Wśród tych celów, domena wiążąca receptor (RBD) białka wypustek COVID-19 (SP) często występuje w repertuarze kandydatów na szczepionki. Jednak immunogenność RBD per se jest ograniczona przez jego niską masę cząsteczkową i strukturalną rearanżację SP o pełnej długości, której towarzyszy oderwanie RBD.

Tutaj pokazujemy, że RBD COVID-19 SP można dogodnie wytworzyć w Escherichia coli po fuzji z fragmentem CRM197, wariantu toksyny błonicy obecnie stosowanej w wielu  skoniugowanych  szczepionkach.

W szczególności pokazujemy, że chimera CRM197-RBD solubilizowana z ciałek inkluzyjnych może zostać ponownie sfałdowana i oczyszczona do stanu obejmującego 5 natywnych wiązań dwusiarczkowych białek macierzystych, zdolność wiązania enzymu konwertującego angiotensynę 2 i zadowalającą stabilność w pomieszczeniu. temperatura. W związku z tym nasze obserwacje dostarczają obowiązkowych informacji do opracowania potencjalnej szczepionki skierowanej przeciwko COVID-19.

Szlak GluN2B-BDNF w jądrze kontaktującym się z płynemowo-rdzeniowym pośredniczy w bólu neuropatycznym w urazem nerwu u szczurów

 

Niniejsze badanie miało na celu zbadanie roli szlaku GluN2B-BDNF w jądrze kontaktującym się z płynem mózgowo-rdzeniowym (CSF-CN) w bólu neuropatycznym.  Do znakowania CSF-CN zastosowano dokomorowe wstrzyknięcie do komory podjednostki B toksyny cholery  sprzężonej z peroksydazą chrzanową (CBHRP). Do obserwowania ekspresji GluN2B i BDNF w CSF-CN zastosowano podwójnie znakowane barwienie immunofluorescencyjne i technikę Western blot. Mannequin szczurzego nerwu kulszowego (CCI) z przewlekłym urazem zaciskowym zastosowano do odtworzenia bólu neuropatycznego.

Oceniano ból w celu określenia działania przeciwbólowego antagonisty GluN2B Ro 25-6981 i przeciwciała neutralizującego BDNF   na szczury CCI. GluN2B i BDNF ulegały ekspresji w CSF-CN, a ich ekspresja była podwyższona u szczurów CCI. Wstrzyknięcie do komory bocznej antagonisty GluN2B Ro 25-6981 lub przeciwciała neutralizującego BDNF w  znacznym stopniu złagodziło przeczulicę termiczną i alodynię  mechaniczną u szczurów CCI. Ponadto zwiększona ekspresja białka BDNF u szczurów CCI została odwrócona przez śródboczne wstrzyknięcie Ro 25-6981 do komory. Wyniki te sugerują, że GluN2B i BDNF ulegają ekspresji w CSF-CN, a zmiana szlaku GluN2B-BDNF w CSF-CN jest zaangażowana w modulację obwodowego bólu neuropatycznego.
